Requirement of FAT and DCHS protocadherins during hypothalamic-pituitary development

چکیده
Pituitary developmental defects lead to partial or complete hormone deficiency and significant health problems. The majority of cases are sporadic unknown cause. We screened 28 patients with pituitary stalk interruption syndrome (PSIS) for mutations in the FAT/DCHS family protocadherins that have high functional redundancy. identified seven variants, four which putatively damaging, FAT2 DCHS2 six recruited through a cohort mostly ectopic posterior gland and/or interruption. All had growth two presented multiple deficiencies small glands. were strongly expressed mesenchyme surrounding normal developing human pituitary. analyzed Dchs2-/- mouse mutants anterior hypoplasia partially penetrant infundibular defects. Overlapping abnormalities distinct morphogenesis observed Fat4-/- Dchs1-/- but all animal models displayed commitment cell type. Together our data implicate hypothalamic-pituitary development identify as candidates underlying such

Hypothalamic-Pituitary-Adrenal Axis Programming after Recurrent Hypoglycemia during Development
Permanent brain injury is a complication of recurrent hypoglycemia during development. Recurrent hypoglycemia also has adverse consequences on the neuroendocrine system. Hypoglycemia-associated autonomic failure, characterized by ineffective glucose counterregulation during hypoglycemia, is well described in children and adults on insulin therapy for diabetes mellitus. Whether recurrent hypogly...

The hypothalamic-pituitary axis is recognized to an increasing extent as a physiological unit which influences and is influenced by endocrine and autonomic function. Anatomically, however, a clear distinction is to be drawn, with regard to both cellular and vascular anatomy, betweenthe neural (hypothalamus and neurohypophysis) and the glandular epithelial (pars distalis) components of the axis....

Professor P. M. Daniel has described in the first paper the nervous pathways connecting the hypothalamus to the posterior pituitary and how the posterior pituitary merely stores the hormones vasopressin, oxytocin (which has no clearly defined function in man), and neurophysin, the large molecule or molecules which bind the other hormones. For many years the anterior pituitary was thought to be ...
متن کاملAdaptation of the hypothalamic-pituitary hormones during intensive endurance training.
OBJECTIVE Physical activity leads to changes in the hypothalamic-pituitary hormonal system. However, acute and long-term adaptations have not yet been precisely characterized. In this study, the changes of the hormonal system as a result of marathon training and running a marathon were examined. In particular, we focused on adaptations of the hypothalamic-pituitary-adrenocortical (HPA) axis, re...
متن کاملRequirement of cannabinoid receptor type 1 for the basal modulation of hypothalamic-pituitary-adrenal axis function.
The endocannabinoid system affects the neuroendocrine regulation of hormone secretion, including the activity of the hypothalamus-pituitary-adrenal (HPA) axis. However, the mechanisms by which endocannabinoids regulate HPA axis function have remained unclear. Here we demonstrate that mice lacking cannabinoid receptor type 1 (CB1-/-) display a significant dysregulation of the HPA axis. Although ...
